dc.description.abstractThe Eph family of receptor is the largest family of receptor tyrosine kinases known. The ephrins are the membrane tethered ligands for this system. The ephrins are divided into two classes: the glycosylphosphatidylinositol anchored A-class ephrins, and the transmembrane B-class ephrins. Both classes of ephrins are associated with lipid rafts. The ephrins were expressed in MDCK cells, a polarized epithelial cell line that uses lipid rafts in sorting. Ephrin-A5 is apically sorted while ephrin-Bl is basolaterally sorted in this system as judged by immunofluorescence and Western blotting. The C-terminus of ephrin-Bl is not involved in its sorting in MDCK cells. The two classes of ephrins are differentially sensitive to Triton X-IOO and Brij 58, suggesting that they may be localized to different lipid rafts. The presence of ephrins in MDCK cells alters the actin cytoskeleton, which may explain the enhanced response to HGF/SF in this system.en
